检测原理本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人LDLR抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、检测样本和辣根过氧化物酶标记的检测抗体,经过孵育,样本中存在的LDLR与固相抗体和检测抗体结合。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中LDLR的浓度成正比。加入终止液终止反应,在450nm波长(参考波长570 - 630nm)测定吸光度值。

LDLR 分子靶点综述

低密度脂蛋白(LDL)受体(LDLR)是一个由839个氨基酸组成的嵌合蛋白,属于LDLR基因家族。它是一种细胞表面受体,可识别嵌入在LDL颗粒外磷脂层上的ApoB100,还可识别存在于乳糜微粒残余物和VLDL残余物中的ApoE蛋白。LDLR介导富含胆固醇的LDL的内吞作用。LDLR复合物存在于细胞表面的网格蛋白包被小窝(或芽),后者通过衔接蛋白结合LDL-胆固醇,去组装形成网格蛋白包被的内体囊泡,这使LDL-胆固醇在内吞作用过程中发生内在化,还可防止LDL扩散在膜表面。上述过程发生在所有有核细胞中,但是主要发生在肝脏中,通过循环可去除约70%的LDL。上调LDLR导致血脂下降,这可能与甲状腺机能亢进有关,而甲状腺机能减退却相反。
