检测原理	本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人Antithrombin-III抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品和待测样本，经过孵育，样本中存在的Antithrombin-III与固相抗体结合。洗涤去除未结合的物质后，加入生物素化的检测抗体孵育。洗涤去除未结合的生物素化的抗体，加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后，加入显色底物TMB，避光显色。颜色反应的深浅与样本中Antithrombin-III的浓度成正比。加入终止液终止反应，在450nm波长(参考波长570 - 630nm)测定吸光度值。

Antithrombin-III，是丝氨酸蛋白酶抑制剂 Serpin 超家族的成员，由 SerpinC1 基因所编码，研究已发现 Antithrombin-III 是弥散性血管内凝血(DIC)的标志物。该蛋白可以抑制凝血酶和其他活化丝氨酸蛋白酶凝血系统，调节凝血级联反应。Antithrombin-III 是最重要的凝血因子抑制剂，即使 Antithrombin-III 出现微小的变化，其也能显著增加血栓栓塞的风险。Antithrombin-III 可以通过依赖性凝血效应或非依赖性凝血效应来抑制炎症。此外，除了 Antithrombin-III 缺乏症外，Antithrombin-III 及其编码基因 SerpinC1 也可能与许多疾病(如高血压、肾脏疾病)相关。
