检测原理本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性捕获抗体预包被在高亲和力的酶标板上,酶标板孔中预先加入辣根过氧化物酶标记的检测抗体微球。酶标板孔中加入标准品和待测样本,经过孵育,样本中存在的待测物质与捕获抗体和检测抗体结合。洗涤去除未结合的物质后,加入显色底物(TMB),避光显色。颜色反应的深浅与样本中待测物质的浓度成正比。加入终止液终止反应,在450nm波长 (参考波长570 - 630nm) 测定吸光度值。

GRN 分子靶点综述

颗粒体蛋白(GRN)在人类中由GRN基因编码。它的前体蛋白—颗粒蛋白前体,又名畸胎瘤细胞源性生长因子。信号肽被剪切后可产生成熟的GRN,进一步剪切可产生许多分子量为6kDa的活性多肽。肽和完整的GRN都可调节细胞生长。然而,GRN蛋白家族的不同成员可能对细胞生长产生抑制和/或促进作用。GRN家族成员对正常发育、伤口愈合和肿瘤发生有重要作用。GRN可能有细胞因子样活性,可在炎症、创伤修复和组织重塑中发挥作用。人肝吸虫可分泌类似GRN的生长激素,从而导致胆管癌的发展。GRN基因突变可通过高外显率的常染色体显性遗传,产生最高达25%的额颞叶变性。

Used to study Grn-related frontotemporal lobar degeneration with Tdp43 inclusions and nephrogenic diabetes insipidus. Human ortholog(s) of this gene implicated in dementia (multiple); neurodegenerative disease (multiple); neuronal ceroid lipofuscinosis 11; and primary progressive multiple sclerosis. Orthologous to human GRN (granulin precursor).
